摘要
为阐明heregulin-α(HRG-α)对乳腺发育的调控作用及其机理,采用组织培养、毛细管电泳、Western印迹、ELISA等方法,首次对小鼠妊娠期HRG-α对乳腺形态发育、β酪蛋白表达和分泌、Rab3A蛋白表达以及HRG-α诱导的信号转导途径中信号分子的磷酸化状态的影响进行了系统研究.结果表明,在妊娠期,HRG-α能够促进STAT5、p42/p44、p38和PKC的磷酸化以及Rab3A蛋白表达,刺激乳腺上皮细胞的增殖和分化,增加并维持β酪蛋白的表达和分泌.
In order to clarify the biological function of heregulin-α (HRG-α) in the development cycle of mammary gland, the effect of HRG-α on Q-casein expression and secretion, the expression of Rab3A and the mechanism of HRG-α in mammary gland, were detected through capillary electrophoresis, Western blot, ELISA method,etc. The results showed that HRG-α activated STAT5, p42/p44, p38 and PKC and increased Rab3A specifically to induce the proliferation and differentiation of ductal epithelial cells, and to increase and maintain the secretion of β-casein in pregnancy.
