新型免疫抑制剂FK778的研究进展

摘要 FK778是一种从抗风湿药来氟米特的活性代谢产物A771726衍生而来新合成的丙二腈酰胺,它具有与其母体化合物来氟米特同等甚至更高的免疫抑制效能。作为新一代免疫抑制剂的代表,其最重要的优点在于它对T、B淋巴细胞的高效作用,并且抑制动脉壁上平滑肌细胞(SMC)的增殖和迁移,从而推迟微血管病变的发生,减轻慢性排斥反应。同时FK778还可以减少其他免疫抑制剂的剂量,改善长期治疗过程中的药物耐受状况,并减少相关副作用,这使其成为临床器官移植的一个理想药物。
