摘要
目的:建立反相高效液相色谱法,可同时分离分析温莪术油中6种倍半萜类活性成分(莪术二酮、莪术醇、吉马酮、莪术烯、呋喃二烯和β-榄香烯)。方法:采用RP-HPLC法作为莪术油及其制剂倍半萜类活性成分的检测方法,色谱柱为Kroma-sil KR100-5 C_(18)柱(250 mm×4.6 mm,5μm);流动相:甲醇-水(90:10):流速:1 mL·min^(-1);检测波长:215 nm。结果:温莪术油中6种倍半萜类成分在同一色谱条件下得到良好分离。莪术二酮、莪术醇、吉马酮、莪术烯、呋喃二烯和β-榄香烯浓度分别在2.66~26.6,3.47~34.7,5.63~56.3,4.22~42.2,10.2~102,7.44~74.4μg·mL^(-1)范围内与峰面积线性关系良好(r≥0.9997),方法平均回收率分别为98.8%,96.5%,101.2%,105.4%,96.6%,95.2%(RSD<3.0%,n=5)。结论:本方法灵敏、专属,重复性好,且可避免倍半萜类中热敏物质的转化,能够真实、准确地反映温莪术油及其制剂的质量。
Objective:To develop an HPLC for quantitative determination of 6 constituents of sesquiterpenoids in Curcuma wenyujin, including curdione, curcumenol, germacrone, curzerene, furanodiene and β- elemene. Methods: A Kromasil KR100 -5 C18(250 mm×4. 6 mm,5 μm) column was adopted with mobile phase consisted of metha- nol -water (90: 10) at a flow rate of 1.0 mL ·min-1, and the detection wavelength was at 215 nm. Results:This method could simultaneously determinate of six sesquiterpenoids in Curcuma wenyujin, The linear ranges of curdi- one, curcumenol, germacrone, curzerene, furanodiene and 13 - elemene were 2.66 - 26.6,3.47 - 34. 7,5.63 - 56.3,4.22 - 42.2,10. 2 - 102,7.44 - 74. 4 μg · mL- 1 ( r ≥ 0. 9997 ) , respectively. The average recoveries were 98.8% ,96.5%, 101.2%, 105.4% ,96.6% and 95.2% (RSD 〈 3.0% ,n = 5 ), respectively. Conclusion :The RP -HPLC is accurate, rapid and reproducible, and destroying the heat -sensitive compound of sesquiterpenoids may be avoided. It represents the components of essential oil of Curcuma wenyujin and their praeparations literally, so it can used for quality control.
