摘要
目的:对干姜中6-姜酚(Ⅰ)、8-姜酚(Ⅱ)和10-姜酚(Ⅲ)3种姜酚进行含量测定。方法:采用反相高效液相色普法,使用 Agilent Hc-C_8柱(150 mm×4.6 mm,5μm),流动相为乙腈-0.1%醋酸水溶液,梯度洗脱(乙腈-0.1%醋酸水溶液比例分别为:0~23 min,43:57:23~50 min,65:35;50~60 min,80:20),流速1.0 mL·min^(-1),检测波长275 nm,柱温30℃。结果:化合物Ⅰ~Ⅲ进样量在0.5~20.0μg范围内与峰面积均有良好的线性关系,相关系数 r 均为0.9999(n=6)。化合物Ⅰ~Ⅲ的平均回收率(n=6)分别为100.6%,101.6%,99.8%;RSD 分别为1.3%,1.6%,2.4%。结论:本方法能够准确测定干姜中3种姜酚的含量。
Objective :To determine three gingerols ,6 - gingerol(Ⅰ) ,8 - gingerol(Ⅱ) and 10 - gingerol(Ⅲ) in Rhizo- ma Zingiberis. Methods:Separation was carried out on Agilent Hc -Cs column (150 mm ×4. 6 mm,5 μm). Solvent A was acetonitrile ,and solvent B was water containing 0. 1% acetic acid. A linear solvent gradient was used ,0 -23 rain, 0 to 43% A;23 -50 rain,to 65% A;50 -60 min,to 80% A. The flow rate was 1.0 mL· min-1 at 30 ℃ ,and the de- tector was set at 275 nm. Results:The linearity was in the range of O. 5 -20 μg for compounds Ⅰ-Ⅲ,and all the cor- relation coefficients were 0. 9999 ( n = 6). The average recoveries ( n = 6) for compound Ⅰ-Ⅲ were 100. 6% ( RSD = 1.3% ), 101.6% (RSD = 1.6% ) and 99. 8% (RSD = 2. 4% ), respectively. Conclusion:This method is accurate, reliable and reproducible for determination of three gingerols in Rhizoma Zingiberis.
