摘要
目的:建立了 HPLC-DAD 同时测定白川降压胶囊中川芎嗪、阿魏酸、欧前胡素和异欧前胡素含量的方法,并与电化学检测进行比较。方法:采用 Dikma C_(18)色谱柱(5μm,150 mm×4.6 mm),甲醇-磷酸盐缓冲液以0.8 mL·min^(-1)线性梯度洗脱。检测波长设置在300,320 nm;电化学检测器的工作电压设为1.2 V 和1.7 V。结果:4种化合物得到了较好的分离,其浓度分别在1.75×10^(-4)~8.75×10^(-3)mg·mL^(-1),1.49×10^(-4)~7.47×10^(-3)mg·mL^(-1),4.06×10^(-4)~2.03×10^(-2)mg·mL^(-1),3.86×10^(-4)-1.93×10^(-2)mg·mL^(-1)范围内线性关系良好。结论:该方法快速、准确,可用于此种新药常规的质量控制。
Objective:An HPLC method with diode array detection (DAD) was developed and validated for simul- taneous determining ligustrazine, ferulic acid, imperatorin and isoimperatorin in Baichuan Jiangya capsules, and it was compared with HPLC - electrochemical detection (ECD). Method:HPLC separations were carried out on a Dikma CI8 column (5μm,150 mm× 4. 6 mm) at the flow rate ofO. 8 mL· min-1 with a linear gradient elution system using methanol and phosphate solution as eluent. Detection was performed using the DAD at 300 and 320 nm,and the potential of working electrode was set at 1.2 V and 1.7 V. Results:Good linear relationships between the peak area and concentration obtained by DAD were found in the ranges of 1.75 ×10-4- 8.75 ×10-3 mg · mL-1,1.49×10-4- 7.47×10-3mg.mL-1,4.06×10-4- 2.03×10-2mg· mL-1 and 3.86×10-4-1.93×10-2 mg· mL-1 for four compounds, respectively. Conclusion:The analytical method is rapid and accurate. It can be used for routine quality control of this new drug.
