摘要 为探讨PGN、LPS和Poly I:C刺激对过敏性哮喘患者嗜碱粒细胞释放能力的比较,并了解患者嗜碱粒细胞上Toll样受体(TLR)的活性,分别用PGN、LPS和Poly I:C刺激患者外周血嗜碱粒细胞,观察其脱颗粒指数(DI)、特异性IgE以及与嗜碱粒细胞表面IgE受体的关系,同时检测了患者过敏原皮试、特异性IgE和尘螨刺激的嗜碱粒细胞DI。结果表明:PGN对患者嗜碱粒细胞的脱颗粒作用强于LPS和Poly I:C[DI分别为(40.7±10.9)%对(19.6±8.9)%、(22.5±7.9)%,P<0.01],同时对患者嗜碱粒细胞的脱颗粒作用也强于正常对照组[(40.7±10.9%)对(15.2±9.0%,P<0.01)]。PGN刺激与粉尘螨刺激DI不相关(P>0.05)。PGN可增强过敏性哮喘患者嗜碱粒细胞的释放能力,其嗜碱粒细胞上存在着相应的TLR。 To study the effect of PGN, LPS or Poly I:C on basophils release-ability and to find out the activity of Toll-like receptor (TLR) on the basophils in patients with allergic asthma. The PGN, LPS and Poly I:C was used to stimulate the peripheral basophils in patients and normal subjects respectively. The Basophil degranulation was observed. Allergen skin test, specific IgE as well as basophil release-ability stimulated by mite were observed to analyze the relationship between the specific IgE antibody and IgE receptors on basophil. The results showed that basophil degranulation stimulated by PGN in allergic asthma patients was much higher than that of by LPS and Poly I:C (P 〈0.01 ), and was also significantly higher than that of in normal subjects ( P 〈 0.01 ). Basophil release-ability stimulated by PGN was not related to that stimulated by mites ( P 〉 0.05 ). The PGN can increase basophil degranulation in allergic asthmatics. TLR exists on basophils in allergic asthma patients.
