摘要
本文介绍了一种快速高效液相色谱法定量测定玉米中真菌毒素玉米赤霉烯酮(Zearalenone,ZEA)的方法。样品用甲醇、乙腈、水混合液提取,提取液经液-液分配萃取、硅镁吸附剂柱层析纯化后,用反相柱ODS-Hypersil分离,流动相为甲醇或者甲醇与水混合液。经实验证实:对ZEA的定量测定用荧光检测法或紫外检测法均有效,该方法对ZEA在0.4μg/g水平上添加的回收率为78.6%—85.3%,平均值为(81.2±7.9)%,当使用荧光检测时,该方法最低检测限为:0.01mg/kg。
A rapid procedure was established for determination of mycotoxin zearalenone residue in maize by high performance liquid chromatography. The sample was extracted with a mixture of methanol, acetonitrile and water. The extracts was cleaned up by both liquid_liquid partitioning and column chromatography. A reversed phase HPLC with fluorescence detector or ultraviolet detector was utilized for both qualification and quantitation. The recoveries for samples fortified with 0.4 mg/kg zearalenone was 81.2± 7. 9%, the minimum detection limit of this method was 0.01 mg/kg.
