摘要
目的:探讨茜草醇提物(RCL)抗肿瘤作用及机制。方法:检测RCL的抑瘤作用;血细胞分析仪对小鼠红细胞数(RBC)、白细胞数(WBC)和血小板(PLT)进行检测,全自动生化分析仪对小鼠血清的谷草转氨酶(AST)、谷丙转氨酶(ALT)、尿素氮(BUN)和肌酐(Cr)水平进行测定;流式细胞术检测细胞周期和细胞凋亡。结果:RCL灌服剂量为500 mg/kg和1000 mg/kg能不同程度抑制小鼠U14宫颈癌实体瘤的生长和延长U14小鼠的生存时间,肿瘤抑制率分别是47.68%和58.94%,生命延长率分别为47.92%和76.70%;阻止肿瘤细胞于G1期;RCL对小鼠造血系统、肝功和肾功没有影响。结论:RCL对小鼠U14宫颈癌具有一定抑制作用,其作用机制可能与抑制肿瘤细胞周期有关。
Objective: Effect of RCL on anti--tumor and mechanism. Method:The anti--tumor effect of RCL was tested, the number of red blood cells (RBC) in mice,WBC and platetet (PLT) were tested by hema- tology analyzer,the serum aspartate aminotransferase (AST),alanine aminotransferase (ALT), urea Nitrogen (BUN) and creatinine (Cr) levels were determined by fully automatic biochemical analyzer; cell cycle and apoptosis were analyzed by flow eytometry. Resuits:RCL (500mg/kg and 1000mg/kg) could inhibit mice bearing in U14 solid tumor growth and prolong the survival time,the inhibition rate up to 47. 68% and 58. 94%, increasing rate in life to 47. 92% and 76.70%. RCL could stop tumor cell in G1 phase without hematopoietic system, liver and kidney injury. Conclusion:RCL has some inhibition effect on mice bearing in U14 cervical cancer and its mechanism might be related to tumor cell cycle inhibited.
