摘要
目的通过观察帕金森病(PD)大鼠在不同时间点脑组织中伊文斯蓝(EB)含量变化来探讨急性和慢性PD大鼠血脑屏障(BBB)通透性的变化。方法脑立体定向将6-羟基多巴胺(6-OHDA)注入大鼠右侧纹状体建立PD模型。将大鼠随机分成对照组和模型组,后者又分为6 h,12 h,24 h,4周组,最后一组为慢性PD组。按BELAYEV法使用EB测定BBB通透性。结果急性PD大鼠BBB通透性在6 h达到高峰(P<0.01),12 h逐渐下降(P<0.05),持续至24 h(P<0.05),慢性PD大鼠(4周)与对照组相比无明显差异(P>0.05)。结论急性PD时伴有BBB的通透性增加,而慢性PD时BBB的通透性未见改变。
Objective To study the alteration of blood - brain barrier (BBB) with parkinson disease (PD) measured by EB content in the brain tissue of rats with acute and chronic parkinson disease. Methods The parkinson disease model was established by direct injection of 6 - OHDA into the right stfiatum area. The rats were randomly divided into control group and model group. The rats in model group were killed at hour 6, hour 12, hour 24 and week 4. The last group was chronic parkinson Disease rat model. The BBB permeability was assessed by Belyaevg method. Results The BBB permeability reached the peak at hour 6 ( P 〈 0.01 ) after 6 - OHDA acute lesion, then decreased at hour 12 ( P 〈 0.05 ), lasted until 24 h( P 〈 0.05 ). The BBB permeability of chronic parkinson disease rats had no significant difference as compared with control group( P 〉0.05 ). Conclusion The permeability of BBB will increase in acute parkinson disease rats but has no change in chronicity parkinson disease rats.
