海洋来源的口蛄虾生物碱的分离纯化和抗鼻咽癌作用研究 被引量:9

Separation and Purification of Alkaloid from Marine Oratosquilla and its Anti-Nasopharyngeal Carcinoma Effect

摘要 目的从海洋来源的口蛄虾中提取并纯化该生物碱,研究其抗鼻咽癌作用。方法系统溶剂法初步分离口蛄虾乙醇粗提物,大孔树脂HP20层析分离正丁醇洗脱的组分;对富含生物碱的组分合并后萃取总生物碱;自动层析仪纯化总生物碱,254 nm波长检测,手动收集洗脱液;质谱仪分析生物碱分子量;MTT方法检测药物对细胞的生长抑制率,绘制细胞生长曲线。结果大孔树脂HP20能非常有效地富集分离的生物碱,质谱分析纯化的生物碱D3分子量为234.33。分离纯化的生物碱对鼻咽癌细胞增殖具有明显的抑制作用。结论口蛄虾来源的生物碱分子量为234.33,对鼻咽癌细胞的增殖具有明显的抑制作用,并呈时间-剂量关系。 Objective To extract effective alkaloid from marine Oratosquilla materials and to study its effect on Nasopharyngeal Carcinoma cell proliferation and purify. Methods Ethanol extract from marine Oratosquilla was diluted by system solvent. Macroporous absorption resin HP20 column chromatography was used to separate n - Butanol diluted group and total alkaloids were extracted. Auto- chromatography was employed to purify alkaloid. The purified alkaloid molecular weight was analyzed by Mass Spectrometry. MTF method was employed to investigate the inhibitory rate on cells proliferation, and growth curves were protrated at the same time. Results Alkaloids were mainly segregated in HP20 chromatographic diluted groups. Purified alkaloid D3 was obtained through Auto - chromatography method. Molecular weight of the purified alkaloid was 234.33 by Mass Spectrometry technique. The alkaloid D3 had obvious inhibitory effects against both CNE1 and CNE -2Z cells. Conclusion The marine -derived alkaloid D3 has prominent anti - cancer cell proliferation effects in a time - and dose - manners, and its molecular weight is 234.33.
