摘要
目的分析不同产地丹参的遗传背景与遗传关系,为丹参药材的栽培育种提供依据。方法采用RAPD与ISSR标记对4个产地的野生丹参、6个产地的栽培丹参共计50个样本进行分析,利用分析软件计算遗传相似性,建立遗传聚类图。结果DNA标记共检测了102个位点,多态条带比率(P)为95.10%,栽培丹参的P值高于野生丹参;居群的总遗传变异H t为0.238 9;UPGMA聚类可以将不同来源的丹参很好地区分开来。结论不同产地间的丹参存在丰富的遗传多样性,丹参种质在遗传背景上较为复杂,不同产地的丹参已出现明显的遗传分化,但是与地理分布没有相关性。
Objective To study the genetic background and relationship of Salvia miltiorrhizae from different population, and provide reference for the seed selection and cultivation. Methods Genetic relationship of total 4 wild populations and 6 cultivated populations of S. miltiorrhizae were investigated by ISSR and RAPD analysis, the genetic identity was analyzed by software Popgene 3.2 and the DNA molecular dendrogram was set up by the software NTSYSpc 2.1. Results A total of 102 markers with 95.10% polymorphic loci (P) were scored, and the number of P among cultivation population was higher than that of wild population. The Neis gene diversity index (Ht) was 0. 238 9, and the dendrogram from the clustering of UPGMA showed the good classification of all populations. Conclusion S. miltiorrhizae from different population shows abundant genetic diversities, and their genetic background are complex. There are obvious genetic differentiations among different S. miltiorrhizae, but the genetic relationships did not show coherent with geographical distribution.
