摘要 目的:探讨prostasin基因在卵巢癌组织中的表达及意义。方法:①应用4 097靶基因点基因芯片对5例卵巢浆液性腺癌及5例正常卵巢组织进行基因表达谱分析,筛查差异表达基因。②应用RT-PCR检测10例卵巢癌组织、10例卵巢瘤组织及10例正常卵巢组织中prostasin mRNA的表达。结果:①基因芯片筛查出卵巢浆液性腺癌差异表达基因58个,其中表达增高(上调趋势)基因30个,表达降低(下调趋势)基因28个;prostasin基因为表达上调基因(cy5/cy3=3.018)。②prostasin基因在卵巢癌组织中表达显著增高(prostasin/actin:0.415±0.177),与正常卵巢组(prostasin/actin:0.192±40.128)及卵巢瘤组(prostasin/actin:0.237±0.232)比较无显著性差异(P>0.05)。结论:基因芯片可以用于筛查卵巢癌差异表达基因,prosta-sin为表达上调基因,prostasin mRNA在卵巢癌组织中表达显著高于在正常卵巢及卵巢瘤组织中表达。 Objective: To study the expression of prostasin in ovary cancer and its significances. Methods: (1)Gene microarray containing 4 097 genes was used to analyze gene expression patterns in tissue samples from 5 cases of human ovarian serous cystadenocarinoma and 5 eases of normal ovarian. (2)RT - PCR method was used to investigate the expression of prostasin mRNA in tissues samples of 10 eases ovarian cancer, 10 cases benign ovarian tumor and 10 eases normal ovarian. Results: (1)58 genes of differential expression were found in ovarian cancer, the expression of 30 genes increased ( up - regulated), the expression of 28 genes decreased ( down - regulated), prostasin was the up - regulated gene ( Cy5/Cy3 = 3. 018).(2)The level of prostasin mRNA expression in ovary cancer tissues ( prostasin/aetin: 0. 415 ±0. 177) was significantly higher than that in normal ovary tissues (prestasin/aetin: 0. 192 ±0. 128) and behign ovary tumor tissues ( prostasin/aetin: 0. 237 ± 0. 232 ) ( P 〈 0. 05) . Conclusion: Gene microarray could screen differential expression gene in ovary cancer; prostasin is up - regulated gene; the expression of prostasin mRNA is signifieandy higher than that of normal and benign ovary tumor tissues.
