摘要
目的:探讨抑癌基因RIZ1在卵巢上皮性肿瘤组织及卵巢癌细胞系中的表达及其表达变化与卵巢癌发生的关系。方法:以卵巢肿瘤组织及卵巢癌细胞系为研究对象。RT-PCR检测RIZ1 mRNA水平;蛋白印迹检测RIZ1蛋白表达。结果:卵巢癌组织、卵巢癌细胞系、良性肿瘤组织及正常卵巢组织中RIZ1 mRNA相对含量的平均值分别为0.42±0.16、0.34±0.12、0.68±0.12及1.17±0.08,RIZ1蛋白相对含量的平均值分别为0.48±0.11、0.38±0.08、0.82±0.11及1.56±0.14。卵巢癌组织、卵巢癌细胞系、良性卵巢肿瘤组织分别与正常卵巢组织比较,RIZ1基因的表达差异均有统计学意义(P<0.05);良性卵巢肿瘤组织与卵巢癌组织比较,RIZ1基因的表达差异有统计学意义(P<0.05)。结论:RIZ1基因表达缺陷与上皮性卵巢癌的发生密切相关。
Objective: To explore the expression of RIZ1 gene, evaluate the clinical significance of RIZI gene expression defect in epithelial ovarian tumors. Methods: The expression of RIZ1 mRNA was detected by RT - PCR. RIZ1 proptein was examined by Western blot in 31 cases of primary epithelial malignant ovarian tumors, 15 cases of epithelial benign ovarian tumors, 5 cases of ovarian cancer cell lines and 12 cases of normal ovarian tissues. Results: The expression of RIZ1 mRNA and protein decreased significantly in all malignant ovarian tumor tissues, ovarian cancer cell lines and benign ovarian tumors. Compared with normal ovarian tissues, the expression of RIZ1 gene decreased significantly in malignant ovarian tumor tissues and ovarian cancer cell lines (P 〈0. 05). The expression of RIZ1 gene in malignant ovarian tumor tissues was significantly lower than that in benign ovarian tumors. Conclusion: Defect of RIZ1 gene expression plays an important role in the development of epithelial ovarian tumors.
