z-VAD-fmk对吸入性损伤小鼠肺脏细胞凋亡及Bcl-2和Bax表达的影响

摘要 目的:观察广谱caspase抑制剂(z-VAD-fmk)对吸入性损伤小鼠肺脏细胞凋亡及Bcl-2和Bax表达的影响。方法:72只BALB/c小鼠造模后随机分为对照组与治疗组各36只,每组设2、6、12、24、48、72 h共6个相应时段,每时段6只,治疗组腹腔给予z-VAD-fmk 6mg/kg,对照组用生理盐水代替,分别于上述各时段剖胸取肺,行常规病理切片并用免疫组化方法观察Bcl-2和Bax表达的变化,计算各时相点肺脏细胞凋亡指数(AI)。结果:小鼠吸入性损伤6 h后,与对照组相比,治疗组AI降低,Bax表达减弱,Bcl-2表达增强,差异具有统计学意义(P<0.05),12 h后差异具有显著性(P<0.01)。结论:小鼠吸入性损伤后,z-VAD-fmk可以增强Bcl-2表达及减弱Bax表达,抑制肺脏细胞凋亡。 Objective: To observe the effect of caspase inhibitor (z-VAD-fmk)on apoptosis and expression of Bcl-2 and Bax in mice lungs with inhalation injury. Methods: 72 BALB/c mice were randomly divided into control group and treatment group after inhalation injury, 6 time points of 2, 6,12, 24, 48 and 72 h were set in two groups separately.The treatment group were given z-VAD-fmk 6mg/kg by intraperitoneal injection. For the control group, 0.9% saline was applied using the same method in treatment group.The mice were killed and the lungs were immediately removed separately at the time points mentioned above. The expression of Bcl-2 and Bax were observed by immunohistochemical method and apoptosis index (AI) was calculated. Results:Compared with control guoup,the AI and expression of Bax decreased and Bel-2 increased in treatmeng group after 6 h and had statistical differences (P〈0.05),the result after 12 h had significant difference(P〈0.01 ). Conclusion: Z-VAD-fmk can increase the expression of Bcl-2 and decrease the AI and expression of Bax after mice with inhalation injury.
