摘要
目的:探讨FTY720体外抑制人胰腺癌Miopac2细胞株增殖的作用。方法:MTT比色法观察FTY720对Miopac2细胞体外增殖的抑制作用;流式细胞仪检测FTY720对Miopac2细胞的细胞周期、凋亡率的影响;倒置显微镜观察细胞形态学变化。结果:随着FTY720浓度的增加,MTT实验中Miopac2细胞吸光度逐渐降低。流式细胞仪分析显示FTY720将Miopac2细胞阻滞于G1期,并使Miopac2细胞发生凋亡,且凋亡率呈浓度依赖性。倒置显微镜下观察有凋亡细胞存在。结论:FTY720对Miopac2细胞的生长有明显的抑制作用,且这种抑制作用具有浓度依赖性。其抑制效应与阻滞肿瘤细胞的细胞周期和诱导肿瘤细胞凋亡有关。表明FTY720具有一定的抗肿瘤作用。
Objective: To investigate the proliferation inhibition effect of FTY720 on human pancreatic cancer Miopac2 cell strain in vitro. Methods: MTT assay was used to measured the proliferative inhibition effect of FTY720.Flow cytometry was used to detect the effect of FTY720 on cell cycle and apoptotic rate. Morphologic changes were observed by invert microscope. Results: The light absorption value of Miopac2 cells decreased with increasing of the concentration of FTY720 in experinaental groups in MTT test. The result of flow cytometry demonstrated that FTY720 induced Miopac2 cells arrest in G1 phase and apoptosis with dose dependent. The apoptotic cells were found by microscopy. Conclusion: FTY720 inhibits the growth of Miopac2 cells with concentration dependent, which may be correlated to cell cycle arrest and cell apoptosis. It suggests that FTY720 has an antitumor effect.
