IP-10对变应性接触性皮炎小鼠Th1/Th2型细胞因子表达的影响 被引量:5

摘要 目的:探讨干扰素诱导蛋白10(IP-10)对变应性接触性皮炎(ACD)小鼠体外培养淋巴细胞分泌Thl/Th2型细胞因子表达的影响,进一步探讨变应性接触性皮炎的发病机制。方法:建立ACD小鼠模型,分离纯化小鼠脾淋巴细胞,并和不同浓度的IP-10共培养,RT-PCR法检测与IP-10共同培养的小鼠脾淋巴细胞中IFN-γ及IL-4 mRNA水平,2-△△CT法分析结果。结果:IP-10高、中浓度组体外培养的ACD小鼠淋巴细胞IFN-γmRNA的表达明显高于对照组,差异具有统计学意义(P<0.01和P<0.05),IP-10低浓度组体外培养的ACD小鼠淋巴细胞IFN-γmRNA的表达与对照组比较差异无统计学意义(P>0.05)。IP-10 3个浓度组之间比较,IFN-γmRNA表达差异均具有统计学意义(P<0.01和P<0.05);IP-10 3个浓度组之间及与对照组比较,体外培养的ACD小鼠淋巴细胞IL-4 mRNA的表达差异均无统计学意义(P﹥0.05)。结论:IP-10可诱导IFN-γ生成增多,促使T细胞向Th1细胞趋化,使Thl/Th2分化失衡,促进了变应性接触性皮炎的发生发展。 Objective: To study the function of interferon-inducible protein-10 (IP-10) in ACD through observing its effect on Th1/Th2 cytokines. Methods: The ACD mouse model was established with 2,4- dinitrochlorobenzene (DNCB).After the separation and purification of spleen lymphoeytes from mice, spleen lymphocyte was co-cultured with IP-10.RT-RCR was used to detect the mRNA of IFN-γand IL-4. 2^-△△CT method was used to analyze the difference of IFN-γand IL-4 mRNA among the four groups. Results: Compared with the control group, 20 and 100 ng/ml concentration groups of IP-10 could remarkably promote the expression of IFN-γin vitro culture CD4^+T cell of ACD mouse (P 〈0.01, P 〈0.05),while there was no significant difference between the 10 ng/ml concentration group of IP-10 and control group. There was significant difference among three different concentration groups of IP-10 in their promoting the expression of IFN-γin vitro culture CD4^+ T cell (P 〈 0.01 ,P 〈 0.05);There was no significant difference among the three different concentration groups of IP-10 on the expression of IL-4 in vitro culture CD4^+T cell, as well as when they compared with the control group (P 〉 0.05). Conclusion: IP-10 may play an important role in the imbalance between Thl and Th2 by inducing the expression of IFN-γ which promotes the development of ACD.
