摘要
目的研究高浓度葡萄糖对蛋白非酶糖化的影响以及药物对蛋白质非酶糖化的干预。方法小牛血清白蛋白(40g/L)与不同浓度的葡萄糖(0.1%、0.5%、1.5%、4.5%)体外孵育1、3、7、14、21、28d,小牛血清白蛋白(40g/L)和1.5%葡萄糖液中分别加入不同浓度的盐酸氨基胍或二甲双胍(5、25、50、100mmol/L)孵育1、14、21d。用NBT还原法测定蛋白质早期糖化产物。结果糖化蛋白产量与葡萄糖浓度以及孵育时间呈正相关,0.1%、0.5%、1.5%、4.5%的葡萄糖孵育28d后糖化蛋白质产量分别较基值高130%、108%、326%、583%、4.5%葡萄糖孵育28d后糖化蛋白产量分别是0.1%、0.5%、1.5%葡萄糖的8.90、5.36、2.41倍。用氨基胍或二甲双胍(5、25、50、100mmol/L)孵育14d,分别使糖化蛋白质减少到对照组的69%、61%、57%、52%和64%、56%、56%、54%。结论蛋白质非酶糖化与葡萄糖浓度孵育时间呈正相关,氨基胍。
URPOSE To study the protein nonenzymatical glycation in vitro and its inhibition by aminoquanidin or metformin.METHODS The in vitro incubation of buff serum albumin (40g/L) with glucose in different concentration (0.1%, 0.5%, 1.5%, 4.5%) for 1, 3, 7, 14 and 21 days respectively was carried out in vitro. Aminoquanidin hydrochloride (AG) or metformin hydrochloride (MF) in different concentration (5, 25, 50 and 100mmol/L) added to the solution containing albumin (40g/L), 1.5% glucose, then incubated for 1, 3, 7, 14 and 21 days respectively. Early glycated product was measured by NBT reduction method.RESULTS The amount of glycated protein correlated with both glucose concentrations and incubation time. The level of glycated protein incubated in 0.1%, 0.5%, 1.5% and 4.5% glucose for 28 days was 130%, 108%, 326% and 583% higher than the control group, respectively. The amount of glycated protein in 4.5% glucose group was 8.90, 5.36, 2.41 times as those in 0.1%, 0.5% and 1.5% glucose groups respectively when incubated for 28 days. The amount of glycated protein incubated in different concentrations AG and MF (5, 25, 50, 100mmol/L) for 14 days was decreased to 69%, 61%, 57%, 52%, 64%, 56%, 56% and 54% of control group respectively.CONCLUSIONS ① nonenzymatical glycation of protein in vitro is directly proportional to glucose concentration and the incubation time. ② AG and MF may inhibit the protein nonenzymatical glycation.
