摘要
目的观察实验性癫及抗痫时海马生长抑制(somatostatin,SS)mRNA的变化。方法采用电惊厥癫和电刺激耳大神经抗惊厥动物模型,利用原位杂交和计算机显微图像处理技术观察。结果正常组海马下托、CA1-4区和齿状回的多形细胞层有SSmRNA表达,其中以下托、CA4区表达量最多,且在各组的变化最明显;与正常组比较,电惊厥组SSmRNA阳性细胞数显著增多(P<0.05),平均光密度显著增高(P<0.05);与电惊厥组相比,电刺激耳大神经抗惊厥组SSmRNA阳性细胞数无明显差异,平均光密度显著减少(P<0.05);单纯电刺激耳大神经组与正常组比较,SSmRNA阳性细胞数和平均光密度无明显差异。结论电惊厥时,海马SSmRNA表达增强,电刺激耳大神经抗惊厥时SSmRNA表达下降,电刺激耳大神经对电惊厥的抑制作用可能与其在海马不同区域调节SSmRNA的表达有关。
URPOSE To observe the changes of somatostatin (SS) mRNA in the hippocampal neurons following electroconvulsion (ECS) and antioconvulsion by electrostimulation of the great auricular nerve (EGAE).METHODS The epileptic model was induced by ECS. By mean of in situ hybridization and image analy ̄zer for micro-image analysis, the changes of SS mRNA was observed.RESULTS A significant increase in SS mRNA was identified in CA4 area and subiculum following ECS, a significant decrease in SS mRNA was also identified in CA4 area and subiculum following EGAN antioconvulsion.CONCLUSIONS ECS enhances the expression of SS mRNA within the hippocampla CA4 area and subiculum, EGAN antioconvulsion inhibits the expression of SS mRNA. The inhibitory effects of EGAN of ECS is likely relevant to EGAN regulating expression of SS mRNA in hippocampus.
