PTEN和CyclinD1蛋白在结直肠癌组织中的表达及临床意义 被引量:2

CLINICAL STUDY OF RELATIONSHIP BETWEEN EXPRESSION OF PATEN AND CYCLIND1 IN HEPATOCELLULAR CARCINOMA

摘要 目的:研究结直肠癌中的PTEN、CyclinD1蛋白的表达及相互关系,初步探讨它们在结直肠癌的发生发展中的生物学意义。方法:应用免疫组织化学染色快捷法,检测50例结直肠癌组织、16例结直肠正常黏膜中PTEN蛋白和CyclinD1蛋白的表达情况,比较PTEN蛋白表达与临床病理指标的关系,及其与CyclinD1蛋白表达的相关性。结果:(1)结直肠癌癌组织PTEN蛋白表达的阳性率(54.0%)明显弱于正常组织(100%),组间比较差异有显著性(P<0.05),其表达水平与结直肠癌的分化程度、淋巴结转移、浸润深度、Dukes分期有关。(2)结直肠癌组织CyclinD1蛋白表达的阳性率(68.0%)明显高于正常结直肠黏膜组(25.0%),组间比较差异有显著性(P<0.05),其表达随结直肠癌侵润深度增加、淋巴结转移、Duke分期的进展而增高。(3)PTEN蛋白表达强度与CyclinD1蛋白表达强度之间呈负相关(r=-0.547,p<0.05)。结论:结论PTEN蛋白和Cy-clinD1的异常表达在结直肠癌的发生、发展过程中起重要作用。两者同时检测有助于预后判断。 Objective :To study the expression of phosphatase and tensin homology deleted on chromosometen ten (PTEN)and CyclinD1 protein in colorectal cancer,and their correlations with the progression of colorectal cancer. Methods:PTEN and CyelinD1 protein expression were detected by immunohistochemieal method in 50 cases of colorectal cancer,16 cases of colorectal adenomas. PTEN protein expression was compared with clinico-pathologic parameters as related to CyclinD1. Results:1. The positive rate of PTEN was significantly lower in colorectal cancer(54.0%) than in normal tissue(100%) (p〈0.05). The expression of PTEN was associated with the loss of tumor differentiation ,lymph node metastasis ,deep invasion and dukes stage( p〈0.05). 2. The positive rate of CyclinD1 was higher in colorectal cancer(68.0%)than in normal tissues (25.0%)( p 〈 0. 05 ), and with the development of tissue invasiveness, lymph node metastasis and Dukes stage, expression level of CyclinD1 become more higher. 3. There is a significant negative correlation between PTEN and CyclinD1 (r=-0.547, p 〈 0.05). Conclusion: Results suggest that the abnormal expression of PTEN and CyclinD1 may play an important role in the carcinogenesis and progression of colorectal cancer. Detection of both proteins might be helpful in prognosis of colorectal cancer.
