摘要
以核桃品种‘香玲'的成熟种胚为试材,分别以带2片子叶、带1片子叶和不带子叶的胚接入MS培养基上的成苗情况和比较HgCl_2消毒2 min、5 min、NaClO消毒10 min 3种消毒的灭菌效果表明:HgCl_2消毒5 min的效果最佳,1周后的材料污染率仅为7%;采用MS为基本培养基,不附加任何生长调节物质,以不带子叶的胚培养4周后即可成苗,未经过煅炼的苗直接移栽到装有蛭石的营养钵中,其最终成活率可达84.6%。
Mature embryos with two cotyledons, one cotyledon or no cotyledon of walnut cultivar 'Xiang Ling' were used as materials to be inoculated on MS medium substrate in this trial and the sterilization effects were examined by 3 sterilization methods in which the materials were treated with HgCl2 for 2 min, 5 min or NaClO for 10 min. The results showed that: the best sterilization method for materials was HgC12 for 5 min, the contamination rate was only 7% after materials inoculated into MS for one week; mature embryos with no cotyledon seedlings after inoculated into MS without growth regulators for 4 weeks, and transplanted directly into pots with vermiculite as a substrate, the survival rate was up to 84.6%.
