摘要
利用花生白藜芦醇合酶基因特异片段,体外合成带地高辛标记的正、反义RNA探针;并与花生根的组织切片进行RNA原位杂交。结果表明,该基因的转录表达在根的韧皮部,其他组织中未见表达;酵母浸提液处理可使该基因的转录表达明显增强。
Using the specific fragment of Resveratrol Synthase gene of Arachis hypogaea L. as a templet, Digoxigenin-labeled antisense (sense) RNA probe was synthesized through in vitro transcription with T7 (T3) RNA polymerases, and was used to detect specific RS gene expression within tissue sections of peanut roots. In situ hybridization analysis revealed that RS gene transcripts were predominant in phloem. Yeast extract treatment resulted in a significant increase in the gene transcription level.
