摘要 目的测定胎盘滋养细胞中胰岛素样生长因子一Ⅱ(insulin-like growth factor-Ⅱ,IGF-Ⅱ)的表达,评价在妊娠高血压综合征(妊高征)中IGF-Ⅱ对胎盘滋养细胞浸润能力的影响。方法用免疫组化SP法,测定36例正常孕妇、34例妊高征孕妇胎盘组织中IGF-Ⅱ蛋白的表达,并用计算机图像分析系统进行定量分析比较。结果①在两组孕妇的胎盘组织中,其合体滋养细胞、细胞滋养细胞、羊膜绒毛层均能检测到IGF-Ⅱ蛋白的表达,但羊膜绒毛层染色与上述两种细胞相比明显减弱。②妊高征组胎盘中的IGF-Ⅱ与正常组相比,平均光密度显著降低(t=2.528,P<0.05)。③轻、中、重度妊高征胎盘滋养细胞中,IGF-Ⅱ的平均光密度呈现逐渐下降的趋势,但三者之间的差异未见明显统计学意义(P>0.05)。结论在妊娠高血压综合征中,胎盘中IGF-Ⅱ的表达降低可能对滋养细胞的浸润能力产生重要影响。 Objective To investigate the expression of insulin-like growth factor- Ⅱ (IGF- Ⅱ ) in human placenta and to evaluate effect of insulin-like growth factor- Ⅱ on the invasion of placental trophoblast in pregnancy induced hypertension syndrome(PIH). Methods SP immunohistochemical staining method was used to detect the protein of IGF- Ⅱ in placenta of 36 normal cases and 34 PIH cases. The mean density was determined by image analysis technique. Results (1) The positive staining of IGF-Ⅱ was found in the syncytiotrophoblast, cytotrophoblast, amnio-chorion of both normal cases and PIH cases, but less immunoreactivity in the amnio-chorion. (2) The expression level of IGF- Ⅱ in PIH placenta was significantly lower than that of the control group( t = 2. 528, P〈0.05). (3) From mild to severe PIH, the mean density of IGF-Ⅱ decreased gradually,but had no statistical difference (P 〉0.05). Conclusions The lower expression of IGF-Ⅱ may play an important role in the invasion of placental trophoblast in PIH.
