摘要
目的探讨β-catenin和APC蛋白在子宫内膜腺癌中的表达及意义。方法采用免疫组化二步法检测β-catenin和APC蛋白在60例子宫内膜腺癌、20例子宫内膜增生症及30例正常子宫内膜组织中的表达。结果在细胞膜上,子宫内膜腺癌β-catenin阳性表达率(36.67%,22/60)显著低于子宫内膜增生症(70.00%)及正常子宫内膜(63.33%,P<0.05);β-catenin阳性表达率与子宫内膜癌的临床分期、病理分型密切相关;子宫内膜腺癌、子宫内膜增生症可见核表达,表达率分别为(30.00%、60.00%),而正常子宫内膜核表达率为零(P<0.005)。APC蛋白主要定位于细胞核上,在子宫内膜腺癌、子宫内膜增生症及正常子宫内膜组织阳性表达率分别为(63.33%、45.00%、50.00%,P>0.05)。结论在子宫内膜癌发生、发展过程中,β-catenin的改变起着主导作用。
Objective To study the expression and role of β-catenin and APC protein in endometrial adenocarcinoma. Method The expressios of β-catenin and APC protein in 60 cases of endometrial adenoearcinoma, 20 cases of endometrial hypertrophy and 30 cases of normal eutopic endometrial tissue were detected using immunohistochemical staining. Result There was obvious increase in the positive expression rate of β-catenin in endometrial adenocarcinoma (36.67%, 22/60) compared to endometrial hypertrophy (70.00%, 14/20) and normal endometlial tissue (63.33%, 19/30), (P〈0.05); the positive expression rate of β-catenin in endometrial ad significantly correlated with the pathological grade and FIGO stage. The prokaryotie expression was seen in endometrial adenocarcinoma and endometrial hypertrophy and the rate of expression was 30. 00 % (18/60)and 60.00 % (12/20) respectively, but the prokaryotic expression in normal eutopic endometfium was zero (P 〈 0.05). APC protein was mainly located on the nucleus. The positive expression rate in endometrial adenocarcinoma, endometrial hypertrophy and normal eutopic endometrial tissue were 63.33 % (38/60) and 45.00 % (9/20) and 50.00 % (15/30) respectively (P 〉 0.05). Conclusion β-catenin may play an important role, but not APC, in the development and progression of endometrial carcinomas.
