硼替佐米联合柔红霉素对HL-60细胞的凋亡实验

摘要 目的研究蛋白酶体抑制剂Bor单用或联合DNR对HL-60细胞的凋亡作用。方法将不同浓度Bor、DNR及两药联合组和对照组作用于HL-60细胞,采用MTT法测定细胞增殖活性,流式细胞术检测细胞凋亡。结果5nmol/LBor作用24h对HL-60细胞有增殖抑制作用,并诱导其凋亡,随着浓度增加及作用时间的延长可使细胞凋亡率明显增加;10nmol/LBor与1.0μmol/LDNR联合作用72h细胞凋亡率最高,与同剂量两药单独作用相比有显著的差异(P<0.01)。结论Bor能显著抑制HL-60细胞的增殖并诱导其凋亡,Bor联合DNR对HL-60细胞有协同作用。 Objective To investigate the effect ofproteasome inhibitor bortezomib(Bor) alone and combination with daunomycin (DNR) on apoptosis of HL-60 cells.Methods HL-60 cells were treated with Bor, DNR and the combination of them in different concentration. Cell proliferation was analyzed by MTT assay;the apoptosis of HL-60 cells was observed by flow cytometry.Resnlts 5 nmol/1 Bor could effectively inhibit HL-60 cell proliferation and induced its apoptosis after 24 hours.HL-60 cell apoptosis rate significantly increased with time prolongation or does increase.The highest apoptosis rate resulted in 10 nmol/1 Bor combination with 1.0u mom/l DNR after 72 hours ,which show significantly difference (P〈0.01).Conclusion Bor can significantly inhibit the proliferation of HL-60 cells and induce apoptosis, synergistic effectiveness can be found when Bor combined with DNR.
