摘要
以葡萄糖为碳源,逐步增大培养基中丙烯腈浓度进行重复续代培养,从土样中筛选到一株腈水合酶活力较高的菌株.经形态观察和16S,rDNA序列分析,该菌株鉴定为红球菌属(Rhodococcus)菌株,命名为Rhodococcussp.TCCC28001.对菌株TCCC28001培养条件进行了初步研究,确定培养基为葡萄糖15,g/L,酵母粉5,g/L,尿素7,g/L,Co2+10×10-5,mol/L.28,℃,180,r/min培养72,h,腈水合酶活力达892,U/mL.
A strain with nitrile hydratase activity, was isolated and screened from soil, by repeated subcultured in the medium culture containing increasing acrylonitrile and glucose as the carbon source. According to morphology and 16S rDNA sequence analysis, the strain is belonged to genus Rhodococcus, named as Rhodococcus sp. TCCC 28001. After optimizing the culture conditions, the optimum conditions are as follows: glucose 15 g/L, yeast extract 5 g/L,urea 7 g/L, Co^2+ 10×10^-5 mol/L. As the strain was cultured with the optimized culture medium and a rotation rate of 180 r/min for 72 h at 28℃, the nitrile hydratase activity could reach 892 U/mL.
