摘要 目的:分离鉴定大鼠灌胃丹参酚酸B后尿液中的代谢产物。方法:大鼠尿液的醋酸乙酯萃取物用聚酰胺柱色谱和制备HPLC方法分离纯化;根据谱学数据进行结构鉴定。结果:从大鼠尿液醋酸乙酯萃取物中分离鉴定了5个化合物,分别是8-羟基-3,4-二氢喹啉-2(1H)-酮(1),1-甲基-1H-吲哚-2-羧酸(2),反式-3-(3-羟基苯基)丙烯酸(3),1H-吲哚-2-羧酸(4),8-氨基喹啉-2(1H)-酮(5)。结论:化合物1,2,4和5为首次从灌胃丹参酚酸B大鼠尿液中分离得到。 Objective: To study the metabolites of EtOAc layer of the rat urine which was collected after after oral administration of Sal B. Methods: The constituents were isolated by chromatography methods of polyamide and PHPLC. The structure of these compounds were identified by NMR spectroscopic methods. Results: Compounds 1-5 were obtained and their structures were identified as 8-hydroxy-3, 4-dihydroquinolin-2 (1 H) -one( 1 ), 1-methyl-1 H- indole-2-carboxylic acid ( 2 ), (E) -3- (3-hydrox-ylphenyl) acrylic acid ( 3 ), 1 H-indole-2-carboxylic acid ( 4 ), 8-aminoquinolin-2 ( 1 H) -one (5) . Conclusion: Compound 1,2, 4, 5 are isolated from urine of rats after oral administration of Sal B for the first time.
