家蚕丝素固定化果胶酶的研究 被引量:23

摘要 家蚕茧丝经高浓度氯化钙或碱溶液处理后,可制成不同形状的丝素.将这些丝素分别与果胶酶通过吸附以及与戊二醛交联结合,制备了不同形状固定化酶.对固定化酶性质的研究表明:丝素能有效地固定果胶酶;其最适pH值为4.0,比游离酶提高了0.5个单位;最适温度为60℃,比游离酶提高了10℃,改善了果胶酶的适应范围.同时还发现:在制备固定化酶时,戊二醛的浓度以0.25%为好; After the silkworm cocoon was treated by high concentration solution of CaCl2 or alkali, it was utilized to produce different kinds of fibroins (fibroin fiber, fibroin powder and fibroin membrane) which could be as a support material for immobilization of pectate lyase by using glutaraldehyde and the sorption The experiment certified that the concentration of the glutaraldehyde in the immobilization should be controlled at 025%, and the content of the pactate lyase should be regulated at 3 mg/L And it also proved that the optimal pH of the pactate lyase immobilized is 40 and the free enzyme is 35,and the optimal temperature of the pactate lyase immobilized is 60 ℃ and the free enzyme is 50 ℃ Therefore, the fibroin was very useful as immobilized enzyme support material 
