摘要
观察化瘀通络中药—肾络通对实验性大鼠糖尿病肾病(DN)模型肾功能及肾组织结缔组织生长因子(CTGF)mRNA表达的影响。方法SD大鼠40只随机分为正常组、模型组、对照组和中药组,模型组、对照组和中药组应用链脲佐菌素(STZ)60mg/kg一次性腹腔注射制备大鼠糖尿病模型。中药组给予肾络通煎剂灌胃25g/(kg·d),对照组给予贝那普利水溶液10mg/(kg·d),正常组和模型组给予等量0.9%氯化钠注射液灌胃。检测各组大鼠24h尿蛋白、血清尿素氮(BUN)、肌酐(Cr)和肾组织CTGF mRNA的表达。结果模型组24h尿蛋白、BUN、Cr和肾组织CTGF mRNA的表达均显著高于正常组(P<0.01);对照组和中药组24h尿蛋白、BUN、Cr和肾组织CTGF mRNA的表达与模型组相比显著降低(P<0.01)。结论化瘀通络中药能够显著减少DN模型大鼠24h尿蛋白,保护DN模型大鼠肾脏功能,并可从基因水平显著降低肾组织CTGF的表达,提示下调CTGF的表达可能是化瘀通络中药保护DN模型大鼠肾功能的重要机制。
Objective To observe effects of dispersing blood stasis and dredging collateral-Shenluotong on renal expression of CTGF mRNA in diabetic nephropathy (DN) rats' kidney.Methods The rats were randomly di- four groups including normal group, model group, control group and Traditional Chinese Medicine (TCM) group. The DN model of rat was made through one - time injection of peritoneal of streptozotocin(STZ) 60 mg/kg.TCM group were intragastric administrated with Shenluotong 25 g/kg· d, Control group and the model group were intragastric administrated with benazepril 10 mg/kg· d, Normal group and model group were given equivalent physiological saline. 24 hour urinary protein, Scr and BUN of different groups, and the expression of CTGF mRNA in nephridial tissue were detected. Results In comparison with normal group,24 hour urinary protein, Scr, BUN and the expression of CTGF mRNA in nephridial tissue of model group were increased obviously ( P 〈 0.01 ) ; While in comparison with model group, those of TCM and Control group were decreaased obviously ( P 〈0.01 ). Conclusion Dispersing blood stasis and dredging collateral can reduce 24 hour urinary protein, protect the renal function and down - regulate the expression of CTGF mRNA in nephridial tissue. It indicated that down - regulation the expression of CTGF maybe an important way of disperse blood stasis and dredge collateral protect rats' renal function.
