摘要
目的探讨乳腺癌耐药相关蛋白(ABCG2)、缺氧诱导因子1α(HIF-1α)蛋白的表达与乳腺癌病理特征及预后之间的关系。方法选取122例接受乳腺癌手术治疗患者的乳腺癌组织石蜡标本,用免疫组化SP法检测ABCG2、HIF-1α蛋白的表达情况。结果122例乳腺癌组织ABCG2、HIF-1α的阳性表达率分别为40.2%和59.0%。ABCG2表达与肿块大小呈正相关(P=0.018);HIF-1α表达与肿块大小(P<0.01)、淋巴结转移(P<0.01)、临床分期呈正相关(P=0.022)。ABCG2表达与122例患者的无病生存期有负相关趋势,无统计学意义(P=0.052),与106例接受了以蒽环类化疗药为主的辅助化疗患者的无病生存期呈负相关(P=0.033)。HIF-1α表达与122例患者的无病生存期呈负相关(P=0.030)。HIF-1α和ABCG2共表达与122例患者的无病生存期呈负相关,有明显统计学意义(P=0.004)。结论ABCG2表达阳性是接受以蒽环类化疗药为主的辅助化疗的乳腺癌患者的预后不良指标。HIF-1α与ABCG2表达明显相关。HIF-1α与ABCG2共检验可以更好的判断乳腺癌患者预后。
Objective To investigate the expressions of ABCG2 and HIF-lot and their relationship with clinieopathological features and prognosis in human breast cancer. Methods The expressions of ABCG2 and HIF-1α in 122 patients with breast cancer who underwent surgery were determined by immunohistocbemical SP method. Results The positive expression rates of ABCG2 and HIF-1α in 122 patients were 40.2% and 59.0%, respectively. The expression of ABCG2 was positively correlated with tumor size (P 〈0.05 ), and the expression of HIF-1α was positively correlated with tumor size (P 〈0.01 ),lymph node metastasis (P 〈0.01 ),and clinical stage (P 〈0.05). In 122 patients, a negative correlation trend was found between the expression of ABCG2 and disease free survival (DFS), without statistical significance (P =0.052). In 106 patients receiving anthracycline-based adjuvant chemotherapy, the expression of ABCG2 was negatively correlated with DFS (P =0.033). The expression of HIF-1α was negatively correlated with DFS in 122 patients (P =0.030). The coxpression of ABCG2 and HIF-1α was also negatively correlated with DFS in 122 patients (P =0.004). Conclusion The positive expression of ABCG2 suggests poor prognosis in patients receiving anthracycline-based adjuvant chemotherapy after surgery. The expression of ABCG2 is positively correlated with the expression of HIF-1α. Co-detection of ABCG2 and HIF-1α might predict the prognosis better in patients with breast cancer.
