摘要
目的:研究人胶质母细胞瘤(GBM)源性微血管内皮细胞(GBMDMEC)和正常人脑组织源性微血管内皮细胞(GBMDMEC)中Nogo-B的功能表达,了解Nogo-B在GBM血管生成中起的作用.方法:通过显微外科神经手术获得14例GBM标本,利用CD105免疫磁珠内皮细胞分选系统纯化内皮细胞,采用免疫组化染色,Real-time PCR和WesternBlot等方法检测其中Nogo-B在蛋白和mRNA水平上的表达.结果:与正常脑组织源性微血管内皮细胞相比较,Real-time PCR和Western Blot检测结果发现,在GBM源性微血管内皮细胞中Nogo-B表达上调.结论:Nogo-B在恶性GBM中mRNA和蛋白水平上表达均显著上调,证明Nogo-B参与了GBM新生血管形成,与GBM发生发展密切相关.
AIM: To explore the expression of Nogo-B in glioblastoma multiform (GBM) derived microvascular endothelial ceils (GBMDMEC) as well as the possible role in pathogenesis of GBM. METHODS: GBMDMEC and normal brain tissue derived microvascular endothelial cells ( BMEC ) without tumorous changes were obtained from patients undergoing microneurosurgery and purified using a MACS magnetic ceil sorting system with anti- CD105 antibody. The mRNA and protein expression levels of Nogo-B were measured with real-time polymerase chain reaction, Western Blot and immunohistochemistry. RESULTS: Nogo-B mRNA and protein expression levels in GBMDMEC increased greatly compared with the levels in BMEC, and correlated positively with the malignancy of GBM. CONCLUSION: The increasing of Nogo-B expression in GBM is closely correlated to the angiogenesis of tumors and Nogo-B may play an important role in the pathological process of GBM.
