高效液相色谱法测定台农19号菠萝糖分 被引量:13

摘要 以90%乙醇作为提取液,色谱柱为CHO-820钙型阳离子交换柱,柱温为90℃,流动相为超纯水,流速为0.5mL/min,进样体积为10μL,用示差折光检测器检测不同季节成熟的台农19号菠萝果实中的糖类,结果表明:该测定方法的相对标准偏差为0.38%~1.35%,相关系数在0.9998以上,加标回收率为98.34%~100.40%;不同季节成熟的台农19号菠萝果肉的含糖量存在较大差异,夏季果的含糖量最高,秋季果次之,冬季果最低,其中蔗糖含量的季节性差异最为明显。 The determination of carbohydrate was studied by means of high performance liquid chromatography (HPLC). The pineapple pulp was extracted by 90% ethanol solution. The HPLC condition was as follows: CH-820 (Ca) column, ultra-pure water as mobile phase, flow rate of 0.5 mL/min, injection volume of 10 μL, the column temperature at 90℃ with refractive index detector. The results showed that the relative standard deviations range was from 0.38% to 1.35%, the linear related coefficientis more than 0.9998, and the recoveries were from 98.34% to100.46%. The method was applied to isolate and detect the conents of sugars in Tainungl9 pineapple pulp. The results showed that Tainung19 pineapple's carbohydrates content varied in the harvest time with more in summer and least in winter, especially in the sugar content.
