摘要
选取了栽培品种巨峰作为试材,进行了外植体愈伤组织诱导和植株再生的研究.通过葡萄品系巨峰无菌外植体的获得,叶片的愈伤组织诱导、再分化,以器官发生途径实现植株再生.结果表明:外植体消毒灭菌过程中,用1 g/L汞处理7 min,外植体污染率和茎尖褐变的比率适中,而且茎尖被诱导分化的成活率最高,可达30.0%.最适的分化培养基为MS+6-BA1.0mg/L+NAA0.01mg/L;叶片愈伤组织诱导的培养基选择MS附加5.0 mg/L 6-BA和0.5 mg/LNAA较为适宜;MS+6-BA0.5 mg/L+NAA0.05 mg/L最适宜巨峰葡萄愈伤组织再生不定芽;将所得的不定芽继代增值诱导生根,1/2MS+IBA 0.2 mg/L为理想的生根培养基,根长适中,根数较多.初步建立了巨峰葡萄栽培品系的再生体系.
The Kyoho Grapevine was used to study regeneration. Callus were induced from leaves of grapes, differentiated and regenerated into plantlets through organogenesis. The results showed that with 0. 1% HgCl2 to disinfect explants for 7min, the rate of pollution and the rate of browning were moderation, and the survival rate was the highest, which was 30. 0 %. Best medium for the differentiation was MS +6- BA1.0mg/L+NAA0.01mg/L; Leaf callus induction medium of MS additional 5mg/L 6-BA and 0.5 mg/L NAA of leaf callus induction medium was more appropriate; MS+6-BA0.5mg/L+NAA0.05 mg/L was the most suitable for grape adventitious buds callus tissue regeneration; 1/2MS+IBA 0.2 mg/L was the ideal rooting medium, and the root length was moderate, the number of root was large. The initial cultivation regeneration system of Kyoho Grapevines was established.
