胃癌蛋白质组学研究中双向凝胶电泳技术的优化 被引量:2

摘要 目的优化应用于胃癌蛋白质组学研究的双向凝胶电泳技术。方法利用双向凝胶电泳技术分离正常胃黏膜和胃癌组织中的蛋白质。比较不同蛋白提取方法(机械匀浆+TCA沉淀法,碾磨+丙酮/TCA沉淀法)、不同最高聚焦电压、不同样本上样量以及不同pH值范围固相pH梯度胶条对双向凝胶电泳结果的影响。结果机械匀浆+TCA沉淀法得到的蛋白质点较多,但电泳图谱中出现大量横纹,对电泳图后期分析的干扰相对严重;碾磨+丙酮/TCA沉淀法则能达到最高聚焦电压,上样量少,使用pH 5-8固相pH梯度胶条获得可分辨的蛋白质点数最多且效果较好。结论对胃癌组织、正常胃黏膜组织的全蛋白抽提方法和电泳条件进行了优化。
