摘要
用电镜定性观察和体视学定量相结合的方法研究镉中毒小鼠肝细胞线粒体超微结构的变化及硒的保护作用。电镜定性观察发现,镉中毒小鼠肝细胞线粒体极度肿胀,嵴消失乃至空泡化。硒镉联合处理小鼠也有类似的变化,但程度较轻。体视学计量发现Cd处理小鼠肝细胞线粒体平均体积(V)、体密度(VV)、平均外径(D)、面密度(SV)、平均截面积(AX)和平均截面周长(BX)显著高于硒镉联合处理小鼠和正常小鼠(P<0.01),面数密度(NA)、比表面(δX)和数密度(NV)显著低于硒镉联合处理小鼠和对照小鼠(P<0.01);硒镉联合处理小鼠AX、NA和V显著低于对照组(P<0.05,P<0.01),而SV和NV极显著高于对照组(P<0.01),其余参数均与对照组无显著差异。因而认为,镉毒会引起肝细胞线粒体的严重损害,其特点为肿胀、数量减少和异形。
オ stereological analysis with qualitative observation of electron microscope was employed to study the cadmiuminduced injury of hepatic mitochondria in mice and protective effects of selenium.Electronmicroscopic observation showed the severe mitochondrial swelling with lysis of cristae.Similar changes in the hepatocytes of Se and Cd unitedlytreated mice existed but were milder.The morphometric determination of mitochondria indicated that mean volume(V),volume density(VV),mean caliper diameter(D),surface density(SV),mean profile area (AX)and mean perimeter (BX)in Cdtreated mice were significantly higher than those in Se and Cd treated as well as normal mice (P<001).Numerical density,(NV),specific surface(δX)and numerical density on area (NA)were significantly decreased(P<001);NA and V were significantly lower but SV and NV were significantly higher in Se and Cd treated mice than in control mice(P<005,P<001);other values didnt show any significant differences between Se and Cd treated and normal mice.It follows that cadmium caused the severe mitochondiral injury characterized by severe swelling,reduced number and deformation;Se effectively abated the Cdinduced mitochondrial injury in mice.
