多西紫杉醇联合丝裂霉素C抑制宫颈癌细胞的增殖与迁移作用 被引量:1

摘要 目的探讨多西紫杉醇(Doc)联合丝裂霉素C(MMC)抑制宫颈癌细胞的増殖与迁移作用。方法MTT实验测定HeLa细胞増殖能力,损伤修复实验测定细胞迁移速度。计算Q值反映多西紫杉醇和丝裂霉素C联合用药效果。结果多西紫杉醇、丝裂霉素C都能够抑制HeLa细胞的増殖,作用具有浓度依赖性(Doc:0.5μg/L vs 0.1μg/L,P<0.05;MMC:10μg/L vs 2.5μg/L,P<0.05);Q值>1,显示两者联合用药有相加作用;同时损伤修复实验显示联合用药组细胞迁移速度(0.035μm/h)显著低于空白对照组(0.173μm/h,P<0.05)及单独用药组(Doc0.079μm/h,MMC0.075μm/h)。结论多西紫杉醇和丝裂霉素C能抑制宫颈癌细胞増殖与迁移,具有一定量效关系;两者联合用药具有协同作用,这可能与两者药效学靶点不同有关。 Objective To investigate effect of Docetaxel(Doc)combined with Mitomycin C (MMC) on proliferation and migration of HeLa cells. Methods Proliferation and migration ability of HeLa ceils were tested by MTT and wound healing assay respectively. The combined effect of Doc and MMC was evaluated by "Q value". Results The inhibition effect of Doc or MMC on proliferation of HeLa cells was dose dependent (Doc:0. 5 μg/L vs 0. 1 μg/L,P〈0. (15 ;MMC: 10 μg/L vs 2. 5 μg/L,P〈0. 05) ;"Q value"exceed 1.0, which indicated the synergistic effect of Doc and MMC. The migration velocity of cells in (Doc + MMC) group (0. 035μm/h) was lower than Control (0. 173/μm/h,P〈0. 05) group, Doc (0. 079μm/ h) and MMC (0. 075μm/h) group. Conclusion Doe and MMC dose-dependently inhibit proliferation and migration of HeLa cells respectively, and there was synergistic effect of the combination therapy. That may be on account of the different targets of two drugs.
