摘要
成环恒温扩增法是一种新型的核酸扩增方法,它是在具有解链活性的DNA聚合酶的作用下,以新形成的产物为模板进行自身循环扩增的恒温核酸扩增方法。该方法是Notom i等2000年发明的,由于它与其它的核酸扩增方法相比较,具有更快捷,更敏感,更廉价和能即时判断结果等优点,已经应用到了多种病原核酸的检测上。作者结合研制尤氏泰勒虫核酸检测实践对该方法的原理,用途和应用中的优缺点进行了综述。
Loop-mediated isothermal amplification (LAMP) is a novel nucleic acid detection method, which takes place by employing a DNA polymerase with strand displacement activity and a set of four primers that recognize a total of six distinct sequences of the target DNA. This method was invented by Notomi et al. in 2000, and it has been applied in many fields because of its many advantages such as faster, more sensitive, cheaper and easy to judge results etc. over conventional nucleic acid amplification methods. This article reviewed the development and application of the method by taking an example of developing of an LAMP method for detection of Theileria uilenbergi.
