锰处理大鼠PC12细胞的增殖抑制及JNK1/2活化表达的变化 被引量:1

摘要 目的探讨氯化锰(MnCl2)对嗜铬细胞瘤(PC12)细胞增殖的抑制作用及对c—Jun氨基端激酶1/2(JNKI/2)磷酸化活化表达量的变化。方法大鼠PC12细胞在含0—500μmol/L浓度Mncl2的培养基中培养3d后,细胞计数法观察MnCl2对细胞增殖的抑制率。Western blot检测JNK1/2磷酸化及总JNK 1/2表达量。结果MnCh对PC12细胞的增殖有显著的抑制作用,且呈剂量一效应关系。随着MnCh浓度的增加,JNK1/2磷酸化水平逐渐升高,蛋白表达水平分别比对照组升高了1.91、2.86、4.13倍(P〈0.05)。结论.MnCl2对PC12细胞的增殖有显著的抑制作用,而上调p-JNK1/2的表达。 Objective To explore the changes of proliferation and activiaty expression of c-Jun N-terminal kinase1/2(JNK1/2) of pheochromocytoma cells treated by manganese in vitro. Methods PC12 cells were exposed to MnCl2 at the doses of 0 - 500 μmol/L for 3 days. The inhibition of the cells was determined by cell counting, and the perphesphorylation and activity expression of c-Jun N- terminal kinase1/2(JNK1/2) by Western blot. Results MnC12 inhibited the proliferation of PC12 cells in a dose dependent manner. Western blot test showed that MnCl2 increased the perphesphorylation and activity expression of c-Jun N-terminal kinase1/2(JNK1/2) in a dose-dependent manner (P 〈 0.05 ). Conclution The inhibition of the proliferation of PC12 cells induced by MnCl2 could be association with up-regulating p-JNK1/2 expression.
