快速提取白念珠菌总RNA的新方法研究 被引量:2

摘要 目的探讨快速提取理想白念珠菌总RNA的实验方法。方法对静止相和菌丝相的白念珠菌国际标准菌株ATCC90028采用冷酸洗玻璃珠联合Tripure法提取总RNA,用紫外分光光度计测量其OD260、OD280的值,并进行琼脂糖凝胶电泳。结果提取的白念珠菌酵母和菌丝相总RNA产量分别为1.9440、1.9624,且OD260/OD280比值介于1.8~2.0范围内,电泳显示提取的总RNA呈现28srRNA,18srRNA和5srRNA3条清晰的条带。结论冷酸洗玻璃珠联合Tripure新方法快速且完整性好,值得推广。 Objective To investigate the total RNA isolation of C. albicans by fast abstraction. Methods The total RNA of C. albicans ATCC90028 with stationary and hypha phase were isolated by Tripure combined with modified glass beads method. The value of A260 and A280 was measured by ultraviolet spectrophotometer and agarose gel electrophoresis was carried out. Results The output of the total RNA isolation was 1. 9440,1. 9624 respectively and the ratio of OD260/OD280 was between 1.8 and 2.0. The result of electrophoresis showed that there appeared three clear bands-28s rRNA, 18s rRNA and 5s rRNA. Conclusion The modified glass beads combined with Tripure method is rapid with good integration and worthy of spreading.
