摘要
目的探讨食管-胃结合部腺癌的表达基因谱。方法采用基因表达谱芯片Affy-metrix U133A2.0对5例手术切除的食管-胃结合部腺癌及正常胃黏膜标本的基因表达谱进行分析。结果食管-胃结合部腺癌与正常胃黏膜相比较,差异表达基因(2倍以上)共773个。其中,上调的基因有364个,下调的基因有409个。这些相关基因的功能涉及凋亡、细胞信号转导、细胞周期、离子通道及运输、代谢、细胞骨架等方面。结论初步筛选出与食管-胃结合部腺癌相关的多种功能基因,为研究食管-胃结合部癌的发病机制提供了可能候选基因。
Objective To survey the changes of gene expression profiles of adenocarcinoma at esophageal-gastric junction. Methods The gene expression profiles of adenocarcinoma tissues at esophageal-gastric junction taken from 5 patients and of normal gastric mucosa were analyzed by the chip gene Affymetrix U133A 2. 0. Results A total of 773 different expression genes (above 2-fold) was found, including 364 up-regulated genes and 409 down-regulated genes. The fuctions of these related genes were involved in metabolism, cell cycle, apoptosis, cellular signal transduction, cell skeleton and so on. Conclusion The functional related genes of adenocarcinoma at esophageal-gastric junction are screened, which may provide a basis for further exploring its molecular mechanisms.
