应用体外培养模型探讨氟、砷对卵黄囊胎盘结构及功能的影响 被引量:2

摘要 用New建立的体外全胚胎培养模型并结合电镜技术探讨了氟、砷对卵黄囊及胚胎的影响。实验结果表明,随着体外染氟或砷剂量的增高,对卵黄囊及胚胎的损害愈加明显,呈现剂量-反应关系。在联合染毒组,析因分析(3×4)结果表明,氟和砷交互作用的主效应为协同作用,各联合组卵黄囊及胚胎受损害的程度均大于相应的单染氟组和砷组。实验结果提示:卵黄囊超微结构的病理改变是氟或砷所致畸机制之一;氟与砷协同作用的机理与卵黄囊结构和功能受损有一定关联。 Whole embryo culture method of new and electron microscopy were used to detect the damaging effcets of fluoride and arsenic on rat yolk sac (YS) and embryos during gead-fold stage.Resuts showed that rat embryos and YS growing in vitro were harmed with the elevations of concentration of fluoride or arsenic in an apparent dose-respose relationship. It was observed that main effct of interaction between fluoride and arsenic belonged to synergism by means of factorial experiment and analysis(3×4),and that damages of combined groups on YS and embryos were higher than that of single groups. The experimental study suggested that pathological changes of yolk-sac ultrastructures would be one of teratogenic mechanisms of fluoride or arsenic, and that machanism of synergistic effect between fluoride and arsenic would be related to disorders of yolk-sae structure and function.
