摘要
根据共聚焦显微镜ACAS570测量原理,结合巨噬细胞研究,认定性和定量方面,叙述了通过荧光图像测量细胞胞内游离Ca2+、胞浆pH、细胞膜电位、线粒体膜电位、细胞膜磷脂和受体流动性、细胞内蛋白的方法,给出了用于定量测量的荧光探针工作曲线的制定方法和不同实验中选择荧光探针所需注意的问题。
The usage of fluorescence cytometry with confocal microscope (ACAS570) was discussed with some research works on mouse peritoneal macrophages. Different fluorescent probes were described in the usage in qualitative and quantitative measurement on analysis of plasma membrane potenital,mitochondrial transmembrane potential,cytosolic pH,lysosomal pH,cytosolic free Ca2+,mobility of membrane phosphlipid and receptors,and cellular protein content. The procedure of generating a working curve and the principles of choosing suitable fluorescent probes for various measurements were provided.
