医用臭氧局部注射对大鼠受损坐骨神经影响的实验研究

摘要 目的:将医用臭氧注射到大鼠受损坐骨神经周围,观察对大鼠受损坐骨神经功能及病理形态改变的影响。方法:40只SD大鼠建立坐骨神经慢性松结扎损伤(CCI)模型后随机分成5组:Ⅰ模型对照组,Ⅱ臭氧20μg/ml治疗组,Ⅲ臭氧40μg/ml治疗组,Ⅳ臭氧60μg/ml治疗组,Ⅴ臭氧80μg/ml治疗组。分别在术前、术后第1、3、5、7、14、21、28天测定所有大鼠热刺激收缩反应期(TWL),于7、14、21、28天测定所有大鼠坐骨神经功能指数(SFI)及运动神经传导速度(MNCV),于28天处死动物进行形态学观察。结果:造模后第3天开始,大鼠出现热痛敏现象,对照组大鼠TWL差值与术前比较,第3、5、7、14、21、28天均明显增大(P<0.01)。Ⅲ、Ⅳ、Ⅴ组于第14、21、28天时点TWL与对照组相比均明显减小(P<0.01),各组间同一时点SFI、MNCV及光镜下坐骨神经病理形态改变差异无统计学意义。结论:40~80μg/ml臭氧局部注射可能对CCI模型鼠的热痛敏具有一定的治疗作用,但受损坐骨神经功能及病理形态改变无明显改善。 Objective : To investigate the effects of local injection of medical ozone ( 03 ) on injured sciatic nerves. Methods:To establish the rat model of chronic constriction injury(CCI) on sciatic nerves,40 Sprague-Dawley rats were randomly divided into five groups: Ⅰ , control group; Ⅱ , with O3 20μg/ml; Ⅲ with O3 40 μg/ml; Ⅳ, with O3 60 μg/ml; Ⅴ, with O3 80 μg/ml. Thermal withdrawal latency (TWL) of rats were determined by the radiant heat stimulation on preoperation and 1 st, 3rd, 5th, 7th, 14th, 21th, 28th day after operation. The sciatic nerve functional index (SFI)and electrophysiological assessment (MNCV) were measured on 7th, 14th,21 th,28th day. The patho-morph results after killing rats were observed. Results:The allodynia to radiant heat stimulation on the territory of ligated sciatic nerves was found from the 3rd day after operation. There was significant difference (P 〈 0.01 ) in TWL between before and after operation. TWL in Ⅲ, Ⅳ and Ⅴ group was significantly different compared with control group(P 〈0.01 ). The SFI and MNCV were no statistical difference in among groups. Conclusion: Medical ozone by local injection was probably effective for neuropathic pain of rats and it may not bring significant adverse effects on sciatic nerve function and neuropathology.
