摘要 目的探讨血管紧张素Ⅱ(ATⅡ)对胰十二指肠同源盒1(PDX-1)表达的影响及其与胰岛素(Ins)分泌的关系。方法测定RIN-m细胞在基础、ATⅡ及氯沙坦作用下Ins分泌量、细胞内Ins含量及细胞内PDX-1 mRNA和蛋白的表达水平。结果在100nmol/LATⅡ作用下,葡萄糖刺激后Ins分泌量、细胞内Ins含量及PDX-1表达水平均明显降低(P均<0.05)。上述作用可被ATⅡ受体阻滞剂氯沙坦部分逆转。结论抑制PDX-1表达水平可能是ATⅡ影响胰岛β细胞Ins合成及分泌的机制之一。 Objective To study the influence of angiotensin Ⅱ on the expression of PDX-1 and insulin secretion. Methods Basic and glucose-induced insulin secretion in supernate and in RIN-m cells were determined by RIA. The PDX-1 mRNA and protein were detected by in-situ hybridization and immunocytochemistry. Results (1) Pretreatment of the RIN-m cells with angiotensin Ⅱ significantly decreased the levels of glucose-stimulated insulin release, intracellular insulin content and PDX-1 expression. (2) Losartan partly improved PDX-1 expression and insulin secretion. Conclusions The angiotensin Ⅱ-induced impairment of PDX-1 mRNA and protein expressions may be one of the possible mechanisms for Ang Ⅱ to affect the insulin synthesis and secretion.
