摘要
本文研究了芦丁、槲皮素、山萘酚、异鼠李素四种黄酮类化合物在毛细管区带电泳中的迁移行为,考察了缓冲溶液pH值、其离子浓度、有机添加剂、电压、温度等对分离的影响。结果发现在35 mmol/L硼砂缓冲溶液,pH 8.4,10%乙腈作为添加剂,20℃,25 kV条件下,四种黄酮在14 m in内实现了基线分离。利用该方法测定青海产油菜蜂花粉中黄酮苷元含量,结果令人满意。
A simple and rapid method for the separation of four flavonoids by capillary zone electrophoresis has been proposed. To optimize the separation conditions, the background electrolyte eomposition, the pH and concentration of buffer were studied first, and the effects of other key factors such as the voltage and temperature of CE,the kind and eoncentration of additives were also evaluated in this paper. Under the optimum condition(35 mmol/L boric acid running buffer, pH 8.4,20 ℃ ,25 kV voltage, 10% (v/v) of acetonitrile), a satisfactory separation of isorhamnetin, kaempferol, rutin and quercetin could be obtained within 14 min. Under the optimum conditions, flavonoid aglycones in rape bee pollen have been determinated, and the results were satisfactory.
