摘要
目的:建立一种可同时测定常见毒品海洛因(代谢物)、苯丙胺类、氯胺酮的高效液相色谱(HPLC)法,了解吸毒者的毒品使用情况,为司法鉴定提供依据。方法:生物样品采用Strata-X C18固相萃取小柱萃取,反相HPLC法对样品进行分离、分析。色谱柱Phenomenex Luna C18(250 mm×4.6 mm,5μm),流动相Ⅰ:乙腈-30 mmol.L-1NH4AC(10:90,v/v);流动相Ⅱ:乙腈-30 mmol.L-1NH4AC(55:45,v/v),流速为1.0 ml.min-1,检测波长为210 nm。结果:尿样和血浆中6种毒品线性关系良好,最低检测限为1.2-6.0 ng(S/N=3),回收率在98.1%-101.0%之间。结论:所建立的HPLC法操作简便、特异性强、灵敏度高,能满足毒品分析和吸毒者滥用药物监测的需要。
Objective:To develop a HPLC method for simultaneous determination of metabolites of heroin, methamphetamine, MDA, MDMA and ketamine, understand the type of abused drugs and provide evidence for judiciary identification. Methods : Following solid - phase extraction with Strata - X C18 column, chromatography was performed on a Phenomenex Luna C18 (250 mm× 4.6 mm, 5 μm) column, using acetonitrile - 30 mmol· L^- 1 NH4 AC ( 10 : 90, v/v) as mobile Phase Ⅰ , acetonitrile - 30 mmol · L^-1NH4AC(55:45, v/v)as mobile Phase Ⅱ at a flow rate of 1.0 ml · min^-1 and the wavelength was set at 210 nm. Results :The assay exhibited a good linear relationship with the limits of detection( S/ N = 3 ) ranging from 1.2 to 6.0 ng and the recoveries were 98. 1% - 101.0%. Conclusion: The established method is simple, selective and sensitive, which suggests that it be applied for drug analysis and surveillance on drug abuse.
