德氏乳杆菌亚油酸异构酶提取条件的研究 被引量:1

摘要 对德氏乳杆菌中亚油酸异构酶的提取条件进行了研究,并对其粗酶液的性质进行了检测,为下一步的分离纯化工作提供依据。通过实验,确定了超声波破碎产亚油酸异构酶菌体的条件:间歇破碎,每次破碎时间20s,间歇5s,破碎20次。当缓冲液pH值为6.0,菌体质量浓度为0.50g/mL时,以粗酶液的酶活力为指标,确定反应的最佳条件:反应温度为40℃;反应时间为150min;粗酶液与底物比例为1:2。 Extraction conditions of Linoleic Acid Isomerase and properties of crude enzyme was studied in this paper. The optimum disruption condition for Lactobadllus.delbrueckii subsp was: intermittent ultrasonic treatment 20 times and time for each treatment was 20 s, with a 5 s interval. pH of the disruption buffer 6.0, concentration of cell extracted 0.50 g/mL Through determination activity of crude enzyme confirm that reaction temperature 40℃, reaction time 150min, proportion of enzyme and substrate 1:2.
