大孔吸附树脂分离纯化发酵液中α-葡萄糖苷酶抑制剂的研究

摘要 本实验研究了大孔吸附树脂对发酵液中α-葡萄糖苷酶抑制剂的吸附分离特性。选择7种大孔吸附树脂,通过比较其对α-葡萄糖苷酶抑制剂的吸附率和解吸率,筛选出最佳吸附剂,并对其吸附动力学特征及解吸条件进行了研究。结果表明:大孔吸附树脂SZ4对α-葡萄糖苷酶抑制剂有较好的吸附和解吸效果,其吸附的最佳工艺条件为:pH6.0,上样流速为4.5BV/h时,其动态贯穿吸附量为6.3×103U/g树脂。30%乙醇浓度可将α-葡萄糖苷酶抑制剂洗脱下来,解吸率达88%。用大孔吸附树脂SZ4对α-葡萄糖苷酶抑制剂进行吸附分离具有吸附量大、解吸容易、再生方便的特点,十分适合于发酵液中α-葡萄糖苷酶抑制剂的分离纯化。 Seven types of macroporous resin were compared on their adsorption and desorption performances to α-glucosidase inhibitor from fermentation broth of Streptomyces sp. DO406. The optimal resin was selected and its dynamic adsorption charactedsyics and desorpfion conditions were studied. Results showed that SZ4 resin presents the highest adsorption and desolption capacities to the α- glucosidase inhibitor, and its optimal adsorption conditions are: pH 6.0 and flow rote 4.5 BV/h. The maximum adsorption is 6.3 × 10^3 U/ g resin. When 30% ethnal is used as eluant, the desorption rate is 88%. SZ, shows better adsorption and desorption capacity to α- glucosiclase inhibitor, so it is appropriate to separate α-glucosidase inhibitor from fermentation broth.
