加标香肠及水样品中相思子毒素-a高效液相色谱检测 被引量:2

Analysis of Abrin-a from Artificially Contaminated Sausage and Water Samples by High-Performance Liquid Chromatography(HPLC)

摘要 应用高效液相色谱法(HPLC)定性和定量检测加标香肠和水样品中相思子毒素-a(abrin-a)。在固定相为Shodex Protein KW-802.5色谱柱,流动相为0.5mol/L的磷酸盐缓冲液(PB,pH7.3),流速为1mL/min,柱温为25℃,检测波长为280llin等条件下,测定结果:abrin-a保留时间(9.80±O.02)min(n=25);浓度与峰面积呈良好的线性关系,线性范围11.765—941.200μg/ml;最低检测限44ng;加标香肠和水样品中abrin-a回收率分别为104.6%和97.9%。测定结果表明,该方法简便、快速、灵敏度较高、重现性好。 A new high-performance liquid chromatography (HPLC) method emplyed to examine abrin-a from artificially contaminated sausage and water samples was described. Stationary phase applied was Shodex Protein KW-802.5 column, Abrin-a was separated from dopants with the mobile phase 0.5 mol/L phosphate buffered solution (PB, pH 7.3) with a flow rate of 1 mlJmin. The retention time of abrin-a was (9.80 ± 0.02 )min (n = 25 ) . The calibration graph was linear in the range of 11.765- 941.200μg/mL abrin-a. The detection limit was 44 ng. The average recoveries of abrin-a in artificially contaminated sausage and water samples with standard abrin-a were 104.6% and 97.9% respectively. We concluded that this is a simple, sensitive and accurate method.
