他莫西芬对胶质瘤C6细胞垂体瘤转化基因表达的影响

摘要 目的探讨他莫西芬在体外对胶质瘤C6细胞垂体瘤转化基因表达的影响。方法不同浓度(1×10-9、1×10-7和1×10-5mol/L)的TAM在体外作用于胶质瘤C6细胞,逆转录聚合酶链式反应半定量法检测PTTGmRNA的表达,WesternBlot检测其蛋白表达情况。结果RT-PCR显示PTTGmRNA在各组的表达分别为(1.872±0.135)、(1.363±0.138)、(0.973±0.121)、(0.667±0.080),各组间差异有统计学意义,均P<0.01;WesternBlot显示PTTG蛋白在各组的表达分别为(1.961±0.140)、(1.525±0.120)、(0.977±0.116)、(0.687±0.117),各组差异有统计学意义,均P<0.01。结论TAM可抑制PTTGmRNA及其蛋白的表达,且这种效应随剂量的增加而增强。 Objective To investigate the effect of tamoxifen on pituitary tumor transforming gene in glioma cell C6. Methods Glioma cells C6 were treated with different concentrations of tamoxifen. PTTG mRNA was detected by reverse transcription polymerase chain reaction (RT-PCR). The expression of PTTG protein was detected by Western blot assay. Results The expressions of PTTGmRNA and PTTG protein decreased after treated with tamoxifen, there was significantly difference among the groups(P〈 0.05). Conclusion Tamoxifen inhibited the expression of PTTG and the expression of PTTG protein significantly in vitro in dosege-dependent manner.
