非小细胞肺癌患者外周血中调节性T细胞的表达

摘要 目的探讨外周血CD4+CD25high调节性T细胞及其表面膜型TGF-β1的表达与非小细胞肺癌的关系。方法招募非小细胞肺癌患者41人,正常对照42人。采用流式细胞技术分析外周血中CD4+CD25high调节性T细胞数量及膜型TGF-β1的表达。ELISA法测定血清TGF-β1浓度。结果非小细胞肺癌患者外周血CD4+CD25high调节性T细胞数量较正常人明显增多(4.52%±1.61%比2.95%±1.19%,P<0.001),接受化疗后,该群细胞数量明显减少(5.23%±1.41%比3.20%±1.14%,P<0.001),CD4+CD25high调节性T细胞数量与肺癌临床分期及血清TGF-β1浓度呈正相关(r=0.789、0.832,P均<0.001)。肺癌患者外周血CD4+CD25high调节性T细胞表面表达的膜型TGF-β1也较正常人多(18.81%±8.78%比13.78%±4.94%,P=0.002),与临床分期也呈正相关(r=0.705,P<0.001)。结论CD4+CD25high调节性T细胞与非小细胞肺癌关系密切,CD4+CD25high调节性T细胞的数量及其表面的膜型TGF-β1是反应肿瘤临床分期的可靠指标,针对该类细胞的策略将为肿瘤的治疗提供新的思路。 Objective To evaluate the association of CD4^+CD25^high regulatory T cells and membrane-bound TGF-β1 with nonsmall-cell lung cancer. Methods We recruited 41 nonsmall-cell lung cancer patients and 42 healthy volunteers. The expressions of CD4^+CD25^high regulatory T cells and membrane-bound TGF-β1 were detected by flow cytometry. Serum TGF-β1 was measured by enzyme linked immunosorbent assay. Results Higher numbers of peripheral CD4^+CD25^high regulatory Tcells were detected in nonsmall-cell lung cancer when compared to healthy subjects (4.52% ± 1.61% vs. 2.95% ± 1.19%, P 〈 0.001 ). After chemotherapy,this subtype ceils were decreased significantly than before(5.23% ± 1.41% vs. 3.20%± 1.14%, P〈 0.001 ). And it positively was correlated with clinical stage or serum TGF-β1 concentration(r = 0.789 and 0.832, respectively, P〈 0.001 ). Furthermore,the expression of membrane-bound TGF-β1 on the surface of CD4^+CD25^high regulatory T cells in nonsmall-cell lung cancer patients was higher than healthy volunteers(18.81%±8.78% vs. 13.78%±4.94%, P = 0.002), and also showed positive correlation with clinical stage(r = 0.705,P〈0.001). Conclusion CD4^+CD25^high regulatory T cell is closely associated with nonsmall-cell lung cancer, and it along with membrane-bound TGF-β1 might be sensitive marker for clinical stage. Targeting to this subset cells could be clinically useful for tumor therapy.
